    目的 探讨蜂毒肽干预胶原诱导关节炎大鼠模型后对Th17/ Treg 平衡调控及关节炎症的影响?方法 将30 组SD 大鼠随机分为正常对照组6 只,其余24 只SD 大鼠于第1 天尾根部注射牛II 胶原,第10 天再次注射II 胶原建立胶原诱导关节炎模型,第14 天淘汰6 只未建模成功的大鼠,筛选出18 只建模成功的大鼠按随机数字表法分为模型组?蜂毒肽组?甲氨蝶呤组,每组6 只?第14 天蜂毒肽组腹腔注射蜂毒肽(0. 5 mg/ kg,隔日1 次),甲氨蝶呤注射组注射甲氨蝶呤(每只1 mg, 每周1 次,共3 次),模型组?正常组腹腔注射等量灭菌注射用水,第32天麻醉处死后,收集血清,ELISA 法检测血清中TNF-α?IL-10?IL-17 A 浓度,分离大鼠脾脏,提取淋巴细胞,MTS 检测单个核细胞活性,流式细胞术检测Th17?Treg 细胞的比例,HE 染色观察踝关节病理变化?结果 蜂毒肽?甲氨喋呤降低血清中TNF-α?IL-17 A 浓度( P <0. 01, P <0. 05),升高IL-10 浓度( P <0. 05);蜂毒肽?甲氨喋呤抑制淋巴细胞活性( P <0. 05);蜂毒肽?甲氨喋呤能下调Th17 细胞比例( P <0. 05),上调Treg 细胞比例( P <0. 01, P <0. 05);蜂毒肽?甲氨喋呤组减少滑膜增生,抑制炎性细胞浸润,延缓骨和关节软骨破坏?结论 蜂毒肽能调节胶原诱导关节大鼠模型Th17/ Treg 平衡,抑制关节的炎症?

    Abstract:

    Objective To investigate the effects of melittin on Th17/ Treg balance and arthritis-associated inflammation in a rat model of collagen-induced arthritis. Methods Thirty Sprague Dawley rats were randomly allocated into a normal group, while 24 SD rats were injected with bovine collagen II on the 1st day and again on the 10th day to establish a collagen-induced arthritis model. On the 14th day, elimination of 6 unmodeled rats and 18 model rats in which the model had been successfully established were selected and divided into a model group, a melittin group, and a methotrexate group by a random number table method. On the same day, the melittin group was injected with melittin (0.5mg/ kg, once every other day), while the methotrexate group was injected with methotrexate (1 mg only, once per week,three times). The model group and the normal group were intraperitoneally injected with equal amounts of distilled water;then, on the 32nd day, the rats were executed. The concentrations of TNF-α, IL-10, and IL-17 A in serum were determined by ELISA. The spleen of each rat was removed, the leukomonocytes were extracted, the activity of leukomonocytes was detected by MTS, and the proportions of Th17 and Treg cells were determined by flow cytometry.Pathological changes of the ankle joint were also observed by HE staining. Results Melittin and methotrex significantly decreased the concentrations of TNF-α and IL-17 A ( P < 0. 01, P < 0. 05), increased the concentration of IL-10 ( P <0.05), inhibited the activity of leukomonocytes ( P < 0. 05), decreased the proportion of Th17 cells ( P < 0.05), and increased the proportion of Treg cells ( P < 0. 01, P <0. 05). In the melittin group and methotrex group, the proliferation of synovial membrane was decreased, which prevented formation of the pannus and delayed the destruction of bone and articular cartilage. Conclusions Melittin can regulate Th17/ Treg balance and inhibit joint inflammation in rats with collagen-induced arthritis.
