    目的 探讨栀子苷对乳腺炎小鼠乳腺组织及乳腺上皮细胞中IL-6?TNF-α 和IL-1β 表达的影响?方法 将40 只分娩后雌性小鼠随机分为空白对照组?模型组?L 组?M 组和H 组?建模后12 h,L 组给予低剂量栀子苷治疗,M 组给予中剂量栀子苷治疗,H 组给予高剂量栀子苷治疗,空白对照组和模型组给予等量磷酸缓冲盐溶液?建模后24 h 观察干预效果?结果 模型组?L 组?M 组和H 组TNF-α?IL-1β?IL-6 mRNA 水平显著高于空白对照组( P < 0. 05);L 组?M 组和H 组TNF-α?IL-1β?IL-6 mRNA 水平显著低于模型组( P < 0. 05);M 组和H 组TNF-α?IL-1β?IL-6 mRNA 水平显著低于L 组( P < 0. 05);H 组TNF-α?IL-1β?IL-6 mRNA 水平显著低于M 组和L 组( P < 0. 05)?L 组?M 组和H 组TNF-α?IL-1β?IL-6 蛋白水平显著低于模型组( P < 0. 05);M 组和H 组TNF-α?IL-1β?IL-6 蛋白水平显著低于L 组( P < 0. 05);H 组TNF-α?IL-1β?IL-6 蛋白水平显著低于M 组和L 组( P < 0. 05)?结论 栀子苷可抑制脂多糖诱导的乳腺炎症损伤,有降低TNF-α?IL-1β 和IL-6 炎症因子水平的作用?

    Abstract:

    Objective To study the effect of geniposide on expression of IL-6, TNF-α, and IL-1β in mammary glands and mammary epithelial cells of mastitic mice. Methods Forty female mice after childbirth were randomly divided into blank control, model, L, M, and H groups. At 12 h after modeling, the L group was treated with low-dose geniposide, the M group was administered intermediate-dose geniposide, the H group was treated with high-dose geniposide, and blank control and model groups were administered an equal amount of phosphate-buffered saline. At 24 h after modeling, the treatment effects were observed. Results mRNA expressions of TNF-α, IL-1β, and IL-6 in model, L, M, and H groups were significantly higher than that in the blank control group ( P < 0. 05). Expressions of TNF-α,IL-1β, and IL-6 mRNAs in the L, M, and H groups were significantly lower than that in the model group ( P < 0. 05). Expressions of TNF-α, IL-1β, and IL-6 mRNAs in M and H groups were significantly lower than that in the L group ( P <0. 05). mRNA expressions of TNF-α, IL-1β, and IL-6 in the M group were significantly lower than that in M and L groups ( P < 0. 05). The levels of TNF-α, IL-1β, and IL-6 in L, M, and H groups were significantly lower than those in the model group ( P < 0. 05). The levels of TNF-α, IL-1β, and IL-6 in M and H groups were significantly lower than those in the L group ( P < 0. 05). The levels of TNF-α, IL-1β, and IL-6 in the H group were significantly lower than those in M and L groups ( P < 0. 05). Conclusions Geniposide inhibits lipopolysaccharide-induced mammary gland inflammation and reduces the levels of proinflammatory factors TNF-α, IL-1β, and IL-6.
