    目的 探讨miR-34c 在口腔鳞状细胞癌(OSCC)组织及细胞中的表达及其对口腔癌细胞生物学行为的影响?方法 qPCR 检测中国地鼠口腔癌组织中miR-34c 的表达水平,脂质体RNAiMAX 介导miR-34c 模拟物组(miRNA-34c mimics)和miR-34c 抑制物组(miRNA-34c inhibitor)转染Tca8113 细胞,并通过CCK-8 和划痕实验检测调控miRNA-34c 表达对Tca8113 细胞活力及迁移能力的影响?结果 与正常组相比,中国地鼠口腔癌组miRNA-34c 表达水平显著升高?miR-34c 模拟物组Tca8113 细胞增殖能力显著减弱( P < 0. 01),迁移能力降低不明显?miRNA-34c 抑制组miRNA-34c 的表达显著下调,细胞的增殖能力和迁移能力显著增强( P < 0. 05)?结论 上调miRNA-34c 表达能有效抑制口腔癌细胞增殖,迁移能力降低不显著,但有降低迁移能力的趋势,下调miRNA-34c 表达可促进口腔癌细胞的增殖,提高细胞迁移能力?

    Abstract:

    Objective To investigate the expression of miR-34c in oral squamous cell carcinoma (OSCC) and its effect on the biological behaviors of OSCC cells. Methods MiR-34c was detected in the Chinese hamster oral cancer tissue by quantitative real-time PCR. MiR-34c mimics and inhibitor were transfected into Tca8113 cells by Lipofectamine RNAiMAX. Cell viability was measured by CCK-8 assay. Cell migration was determined by a scratch test. Results Compared with the normal group, miRNA-34c expression in the Chinese hamster oral carcinoma group was increased significantly. Upregulation of the miR-34c inhibited the viability of Tca8113 cells ( P <0. 01), but there was no significant change in migration of the Tca8113 cells. Downregulation of miR-34c promoted the viability and migration of Tca8113 cells ( P <0. 05). Conclusions Upregulation of miR-34c effectively suppresses oral cancer cell growth,while non-significantly inhibits the cancer cell migration. Downregulation of miRNA-34c expression promotes proliferation and migration of oral cancer cells.
