    甲基苯丙胺(Methamphetamine,METH)滥用合并人类免疫缺陷病毒(Human immunodeficiency virus,HIV)感染在当今是一个广泛的公共卫生问题。METH与HIV-1 Tat蛋白可单独及协同对神经细胞产生神经毒性,神经细胞受损是造成神经系统损伤的诱因之一,进而是促进HIV相关神经认知障碍(HIV-associated neurocognitive disorders,HAND)发生发展的重要因素。其中小胶质细胞作为中枢神经系统(CNS)中的免疫细胞,生理状态下对中枢神经系统的发育、平衡和先天免疫起着关键作用。在毒性损伤等病理条件下,小胶质细胞可发生显著的病理改变,进而促进神经功能障碍的发生发展。该文综述了近年来METH与HIV-1 Tat蛋白单独及协同对小胶质细胞神经毒性的研究进展,从小胶质细胞病理变化的角度评价这种神经毒性,旨在为毒品滥用合并HIV感染这一特殊人群患HAND后的治疗及干预靶点提供参考依据。

    Abstract:

    The co-abuse of methamphetamine (METH) and human immunodeficiency virus (HIV) infection is a widespread public health issue nowadays. METH and HIV-1 Tat protein can independently and synergistically exert neurotoxicity on nerve cells. The damage of nerve cells is one of the causes of nervous system injury, and it is an important factor promoting the occurrence and development of HIV-associated neurocognitive disorders (HAND). Among them, microglia, as immune cells in the central nervous system (CNS), play a key role in the development, balance and innate immunity of the CNS under physiological conditions. Under pathological conditions such as toxic damage, microglia can undergo significant pathological changes, thereby promoting the occurrence and development of neurological dysfunction. This article reviews the research progress in recent years on the neurotoxicity of METH and HIV-1 Tat protein on microglia independently and synergistically, and evaluates this neurotoxicity from the perspective of microglial pathological changes, aiming to provide reference basis for the treatment and intervention targets of this special population with HAND after drug abuse combined with HIV infection.
