    摘要:

    目的 研究13-甲基十四 烷酸(13-methyltetradecanoicacid, 13-MTD)对大鼠脑皮质星形胶质细胞氧反常的保护作用。方法 原代后传代培养新生SD大鼠乳鼠大脑皮质星形胶质细胞(Astroglia,AST),以氧糖剥夺/再复氧糖(OGD/R)方法复制氧反常模型,OGD 10 h/R 24 h,于再复氧糖即刻分别给予13-MTD 20、40、80 μg/ml(M20、M40、M80)干预,倒置相差显微镜动态观察AST形态,免疫组化鉴定角质纤维酸性蛋白(Glial Fibrillary Acidic Protein,GFAP),MTT法检测线粒体活性,免疫组化法检测AST的AQP4蛋白表达。结果 OGD 10h/R 24h损伤后,体外培养的大鼠乳鼠脑皮质AST出现明显病理损伤,线粒体活性显著下降(P<0.01),AST膜AQP4蛋白表达量明显增加(P<0.01);与模型组比较,13-MTD 20、40、80 μg/ml可减少病理形态损伤,使线粒体活性上升、AQP4蛋白表达减少,以80μg/ml效果最好(P<0.01)。结论 AST OGD 10h/R 24h后,细胞形态改变且线粒体活性降低,AQP4表达升高后细胞发生严重水肿,13-MTD 20、40、80 μg/ml尤其是80 μg/ml可降低AQP4的表达,减轻细胞水肿,进而保护氧反常诱导的AST损伤。

    Abstract:

    Objective To investigate the protective effects of 13-methyltetradecanoic Acid(13-MTD) on focal cerebral ischemia in rats and the oxygen paradox of cultured astrocytes in vitro. Methods Almost pure astrocytes were obtained after Primary culture and subculture at the cerebral cortical of neonatal rats, and then the oxygen-glucose eprivasion 10h/reperfution 24h (OGD10h/R24h) model was made. 13-MTD 20, 40, 80mg/kg was administered immediately after reperfusion. Inverted Phase contrast microscope was used to observe the morphology of cells dynamically. MTT assay was employed to detect the mitochondrial reducibility, and Immunohistochemistry was used to measure the expressions of GFAP and AQP4 Proteins. Results After oxygen paradox, the pathological damage of the cerebral cortical astrocytes in neonatal rats was observed apparently, the mitochondrial reducibility was decreased significantly(P<0.01), the expression of AQP4 protein in AST was increased significantly(P<0.01). Compared with Vehicle, 13-MTD 20、40、80μg/ml can improve the above changes obviously (P<0.01), and it would be better by using 80mg/kg. Conclusions After OGD10h/R24h, the reducibility of astrocytes was decreased. The edema of astrocytes was significantly because of the expression of AQP4 Protein was up-regulated. 13-MTD 20、40、80μg/ml can imProve the above changes obviously and 13-MTD 80 μg/ml would be better. 13-MTD could protect astrocytes after OGD10h/R24h.
