    目的 探索不同EV71毒株在新生鼠的毒力情况。方法 临床轻症、重症和死亡来源的3株EV71毒株,接种1日龄balbc乳鼠,观察14天小鼠临床症状,于感染后1天、3天、5天、7天和9天,处死小鼠取肝、脾、肺、肠、脑和肌肉, RT-QPCR测组织病毒载量, HE染色观察病变。 结果 重症株与轻症株症状严重程度无差异(P=0.693),且两组均较死亡株组重(P=0.000,P=0.000);轻症、重症和死亡株组小鼠生存率分别为77.2%,81.7%和97.8%,差异有统计学意义(P=0.0010, P=0.001,P=0.0004);轻症株组肺出血最重,脑、肌肉、脾和肠病理定性无明显差异;轻症株组各组织病毒量最高,死亡株组最低。结论 临床轻症毒株在乳鼠症状最重,死亡株最轻,与人的临床结局不一致,可能与病毒基因、宿主等因素相关。

    Abstract:

    Objective To explore virulence of enterovirus 71 from infected children in neonatal mouse. Methods Three strains of EV71 were isolated from the mild, severe and dead patients. Symptoms, weight and death of mice were registered throughout 14 days. A group of mice were sacrificed on the first, third, fifth, seventh and nith day post infection to gain tissue like liver, spleen, lung, intestine, brain and muscle tissue which were used to detect the virus tilter by real-time RT-QPCR, and pathological lesions by HE staining. Results As to the severity of symptoms, no significant difference existed between the severe and mild group (P = 0.693), which were more serious than that of the fatal group. (P = 0.000<0.05/6, P = 0.000 <0.05/6).The survival rate of the mice infected with mild, severe and fatal virus was 77.2%, 81.7% and 97.8%, respectively and there was significant difference between the three groups (P = 0.0010 <0.05, P = 0.001 <0.05, P = 0.0004 <0.05). Lung hemorrhage of the mild group was the most serious, and there were no significant differences of brain, muscle, spleen and intestine in pathological lesions. Virus tilter in liver and muscle was higher than the other tissues and that in mild group of different tissues tended to be more than the other two groups. Conclusions Neonatal mouse infected with the mild strains presented the heaviest symptoms, which were not consistent with outcomes of human. It was considered to be related to the virus gene, host and other factors.
