    目的 构建新型长时程抑郁模型(long-term depression model,LTD)小鼠,并对比此种小鼠与常见抑郁小鼠模型的优劣。 方法 通过早期母婴分离,以及断乳后持续慢性应激来构建 LTD 小鼠,对比其与慢性不可预知温和刺激模型(chronic unpredictable wild stress,CUMS)、早期应激模型(early life stress model,ELS)小鼠长期抑郁样行为改变,并对比 eGFP 转基因的 LTD 小鼠与对照小鼠海马神经元变化。 结果 与 CUMS、ELS 小鼠相比,LTD 小鼠在行为学上能更长期的表现出稳定的抑郁状态,组织切片显示 LTD 小鼠海马 CA1 区和 CA3 区呈退行性变性。 结论 成功的构建了 LTD 小鼠,此模型小鼠能导致更长久抑郁样行为,表现出长期的心理应激状态,是一种适合用于长期模拟抑郁状态的模式小鼠

    Abstract:

    Objective To establish a novel long-term depression ( LTD) model in mice, and to compare this model with the common depression mouse model. Methods Using early maternal separation and persistent chronic stress after weaning to prepare LTD mice, which were then compared with chronic unpredictable mild stress and early life stress mice to observe potential differences in their long-term depression-like behaviors. The changes in hippocampal neurons of LTD mice expressing transgenic eGFP were also observed. Results Compared with chronic unpredictable mild stress and early life stress mice, LTD mice showed a stable state of depression in animal behavior experiments. Degeneration of the hippocampal CA1 and CA3 regions of LTD mice was observed in tissue sections. Conclusions The LTD mouse model was successfully established. LTD mice provide a model suitable for the investigation of long-term simulation of depression, which can lead to long-term depression-like behavior and psychological stress.
