      目的 研究杜仲提取液对小鼠乳腺炎的治疗效果和作用途径?方法 通过乳导管灌注LPS 构建小鼠乳腺炎模型,注射LPS 12 h 后通过腹腔进行不同浓度梯度杜仲给药,分为6 组?给药12 h 后对小鼠进行剖杀并取其乳腺组织,利用HE 染色观察各组病理变化,利用ELISA 方法检测乳腺组织中TNF?2α?IL?6?IL?1β 细胞因子的变化情况?结果 1)浓度为50 μL 0. 2 mg/ mL 的LPS 可成功构建小鼠乳腺炎模型;(2)HE 染色结果表明杜仲提取液可减轻LPS 诱导的小鼠乳腺炎的损伤程度,随着注射杜仲提取液剂量的增加,乳腺组织的损伤程度减轻,且注射剂量为20 mg/ kg 和27 mg/ kg 时效果最佳;(3)ELISA 结果表明杜仲提取液可降低乳腺组织中炎性细胞因子IL?1β?IL?6?TNF?α 的含量,且注射剂量为27 mg/ kg 时效果最佳( P < 0. 01)?结论 杜仲提取液可显著减轻小鼠乳腺组织病理损伤?减少乳腺组织内炎性细胞因子IL?1β?IL?6?TNF?α 的释放,阻断了中性粒细胞向乳腺组织和乳腺腺泡腔募集和迁移过程,从而减轻乳腺组织损伤,其中浓度为27 mg/ kg 的杜仲提取液抗炎效果最佳?

      Objective In this study, we tested the therapeutic efficacy and mechanisms of action of Du Zhong injection in a mouse mastitis model. Methods The mastitis mouse model was produced by injection of lipopolysaccharide (LPS) into the breast duct. At 12 h post LPS injection, mice were split into six groups and administered different concentrations of Eucommia ulmoides (Du Zhong) into the abdominal cavity. After a further 12 h recovery, mice were killed and the mammary glands collected. Pathological changes in each group were observed using hematoxylin and eosin staining. Changes in tumour necrosis factor?α(TNF?α), interleukin?6(IL?6) and interleukin?1(IL?1) cytokines in breast tissue were detected by enzyme?linked immunosorbent assay. Results The mouse mastitis model was successfully constructed using 50 μL of 0. 2 mg/ ml LPS. Pathological examination revealed that, increasing doses of Du Zhong injection were associated with decreased mammary gland injury in the model mice, with the greatest effect observed at 20 mg/ kg and 27 mg/ kg. By enzyme?linked immunosorbent assay, Du Zhong injection also reduced pro?inflammatory cytokine expression in the breast tissue, with greatest effects observed at 27 mg/ kg ( P < 0. 01). Conclusions Du Zhong injection can significantly reduce pathological damages, pro?inflammatory cytokine release, and neutrophil invasion in mammary gland tissue in a mouse model of mastitis, with an optimal dosing of 27 mg/ kg.
